Innovative Use of Silver Sulfide Nanoparticles in Light-Responsive Double Network Hydrogels for Cancer Photothermal Therapy

Researchers have recently developed a novel light-responsive double network hydrogel based on methyl acrylated peptide nanofibers (PNFMA) with high biocompatibility, excellent biodegradability, and multifunctionality for the modulation of cancer cells in photothermal therapy.


In their work, researchers achieved the controlled gelation of methyl acrylated peptide and GelMA through photocrosslinking. By using electrostatic interactions to immobilize uniformly dispersed silver sulfide nanoparticles (Ag2S NPs) on the template of methyl acrylated peptide nanofibers (PNFMA), they successfully constructed a light-responsive double network hydrogel (GelMA/PNFMA-Ag2S) via the "smart molecular assembly - light-controlled shaping" strategy. The self-assembly of peptide nanofibers (PNFMA) addressed the issue of low photothermal efficiency due to aggregation while improving biocompatibility.


The hydrogel demonstrated rapid light-controlled gelation properties, with rapid conversion from solution to gel within 2 minutes under 365nm UV light irradiation. Additionally, the hydrogel exhibited excellent photothermal stability both in vitro and in vivo, with a high photothermal conversion efficiency of up to 56.6% and significant efficient ablation of tumor cells. The electrostatically driven Ag2S NPs fixed on PNFMA formed a synergistic nano-bio interface that created a "hotspot-dense" photothermal network, while the double network structure inhibited heat dissipation. As a result, under conditions of 2W/cm2, the tumor ablation rate reached 99.9% with no off-target toxicity.


The incorporation of silver sulfide nanoparticles into a light-responsive double network hydrogel opens up promising possibilities for enhanced cancer photothermal therapy. The unique properties of this hydrogel, including rapid gelation, high photothermal stability, and efficient tumor cell ablation, highlight its potential as a powerful tool in the fight against cancer.


Overall, this innovative approach showcases the potential of combining advanced nanotechnology with biomedical engineering to develop next-generation therapies for cancer treatment.
